ABSTRACT

Triptolide(TP), the main active and toxic component of Tripterygium wilfordii, has the limitations of low bioavailability, poor absorption, low concentration in plasma, and small lethal dose. Microneedle(MN), the hybrid of hypodermic needle and transdermal patch, is a physical penetration-enhancing system. Dissolving microneedles(DMNs) can be tailored to specific needs of degradation rate. In this study, the TP-loaded DMNs(DMNs-TP) were prepared with the two-step centrifugation method. The optimal ratio of PVA to PVP K30, water content in matrix solution, demoulding method, and plasticizer for preparing DMNs were investigated with the indexes of formability and mechanical strength. The drug loading capacity was determined by HPLC and morphological characteristics were observed under an optical microscope. The mechanical properties were investigated by H&E staining and Franz diffusion cell was used to detect the in vitro skin permeation characteristics. Through the experiment, we confirmed that the optimal backing material should be PVA and PVP K30(3∶1) and the optimal ratio of matrix material to water should be 3∶4. The prepared DMNs-TP were pyramidal with smooth surface and length of approximately 550 μm. Each patch(2.75 cm~2) had the drug loading capacity of(153.41±2.29) μg, and TP was located in the upper part of the needle. The results of in vitro skin permeation assay demonstrated that the cumulative penetration of TP in DMNs-TP reached 80% in 24 h, while little TP solution penetrated the skin, which proved that DMNs promoted the transdermal delivery of TP.

ABSTRACT

Delayed-or non-healing wounds caused by trauma, surgical procedures, acute diseases, or chronic diseases, and proli-ferating scar have a serious impact on patients' quality of life and increase the economic and psychological burden on their families. Therefore, how to accelerate wound healing and obtain satisfactory aesthetic results is of great concern to researchers and is an urgent clinical problem to be solved. In recent years, the mechanisms of Chinese medicinal materials in accelerating wound healing and inhi-biting scar formation by regulating cytokines have been clarified, which provides a scientific basis for revealing the efficacy of Chinese medicinal materials against clinical trauma. This review focuses on the therapeutic effects of active ingredients, extracts, and topical preparations of Chinese medicinal materials through regulating cytokines in the inflammation, proliferation, and remodeling phases of wound healing. It is expected to provide evidence for the application of Chinese medicinal materials in wound therapy.

ABSTRACT

OBJECTIVE@#To deeply understand the clinical manifestation, laboratory examination characteristics, diagnosis and treatment of an eight p11 myeloproliferative syndrome (EMS) with rare phenotypes.@*METHODS@#The clinical and laboratory characteristics and the process of allogeneic hematopoietic stem cell transplantation (allo-HSCT) were summarized in 1 rare EMS case involving T/B/myeloid cells. Meanwhile, 2 similar cases in the previous literature were also discussed.@*RESULTS@#The bone marrow examination indicated that the patient with B-cell acute lymphocytic leukemia. The lymph node biopsy showed that the patient was T lymphoblastic/myeloid lymphoma. The 8p11 abnormality was found by the examination of bone marrow chromosomes. The RT-PCR examination showed that the BCR-ABL fused gene was negtive. The FGFR1 breakage was found by using the FISH with FGFR1 probe in lymph node. The Mutation of FMNL3, NBPF1 and RUNX1 genes was found by using the whole exome sequencing. The patient received allo-HSCT under CR2. By the follow-up till to September 2019, the patient survived without the above-mentioned disease.@*CONCLUSION@#EMS manifest as neoplasms involving T-lineage, B-lineage, and myeloid-lineage simultaneously is extremely rare. Although the FGFR1 gene-targeted therapy can be conducted, allo-HSCT should be actively considered.

ABSTRACT

Objective:To study the correlation of eight chemical components in Miao medicine <italic>Periploca forrestii</italic> from different producing areas with the ecological and soil factors. Method:The contents of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, isochlorogenic acid C, procyanidin A<sub>2</sub>, and periplocin were simultaneously determined by ultra-performance liquid chromatography (UPLC). The root soil samples from various producing areas were collected for testing various soil factors, followed by climate data extraction with ArcGIS and topographic data recording using GPS. SPSS 24.0 was employed to conduct the bivariate analysis and stepwise regression analysis of the eight chemical components in <italic>P. forrestii</italic> from different producing areas with the ecological and soil factors. Result:Stepwise regression equations of the content of eight chemical components against ecological and soil factors were established. The findings demonstrated that neochlorogenic acid was negatively correlated with precipitation in the coldest season and chlorogenic acid negatively correlated with precipitation in the driest month. Cryptochlorogenic acid was negatively correlated with precipitation in the coldest season and average temperature in the warmest season, but positively with selenium. Isochlorogenic acid B was mainly affected by soil factors. Specifically, it was positively correlated with available iron and molybdenum but negatively with total phosphorus and available phosphorus. Isochlorogenic acid A was positively correlated with molybdenum but negatively with the coefficient of variation of precipitation. Isochlorogenic acid C showed a positive correlation with exchangeable magnesium. Procyanidin A<sub>2</sub> exhibited a positive correlation with molybdenum and a negative correlation with available potassium. Periplocin was negatively correlated with the coefficient of variation of precipitation. Conclusion:The correlation between the eight chemical components of <italic>P. forrestii</italic> and the ecological and soil factors has been clarified, which will provide reference for the introduction, cultivation, and standardized planting of <italic>P. forrestii </italic>and also a theoretical basis for further research on its ecological and soil factors and quality formation mechanism.

ABSTRACT

Objective:To explore the potential suitable distribution area and the high-quality distribution area of <italic>Sabia parviflora</italic>. Method:Combined with the distribution information and environmental factors,the maximum entropy (MaxEnt) model and ArcGIS software were used to predict the potential suitable distribution area of <italic>S. parviflora</italic>. Based on the correlation between environmental factors and total saponins,total flavonoids,quercetin-3-<italic>O</italic>-gentiobioside,camellianoside,tsubakioside A,kaempferol-3-<italic>O</italic>-rutinoside and isobariclisin-3-<italic>O</italic>-rutinoside,the quality regionalization was conducted by using spatial interpolation method and fuzzy superposition function in ArcGIS software. Result:<italic>S. parviflora</italic> is mainly distributed in Yunnan,Guizhou,Guangxi province in China. The medium and high suitable areas accounts for about 2.88% of the national area. The precipitation in October and November,the precipitation in the warmest and driest seasons,the standard deviation of seasonal changes in temperature and altitude are the main environmental factors that affect the distribution of <italic>S. parviflora</italic>. Slope,precipitation,solar radiation and temperature change had great influence on the accumulation of secondary metabolites. Based on the results of potential suitable distribution and spatial interpolation of each component,the high-quality areas of <italic>S. parviflora</italic> are mainly concentrated in the southwest of Guizhou,with Qinglong,Guanling,Zhenning,Pu'an,Xingren county and other areas as the core. Conclusion:This study provides a scientific guidance for the site selection of artificial planting and the procurement of medicinal materials for <italic>S. parviflora</italic>.

ABSTRACT

<b>Objective::To explore the feasibility of Photoshop image processing software in the micro-spherical diameter measurement and identification of medicinal materials, in order to provide a new method for the measurement and mapping of traditional Chinese medicine. <b>Method::Photoshop (CS3 version and above) software was used to measure the diameter of 200 Notoginseng Radix et Rhizoma albumin microspheres in the same batch, digital microscopy and Photoshop image processing software was used to draw schematic diagrams of microstructures of Notoginseng Radix et Rhizoma medicinal powders and its four common pseudo-products and powders, namely Bletillae Radix, Alpiniae Officinarum Rhizoma, Curcumae Longae Rhizoma and <italic>Manihot esculenta,</italic> and make the identification table for classification and identification. <b>Result::The average diameter of the microspheres was (30.62±4.21)μm, and the diameter was mainly distributed in 20-40 μm. The average diameter of the microspheres was verified by laser particle size analyzer (30.18±4.67)μm. The differences between the two methods were not statistically significant. There were many starch granules in Notoginseng Radix et Rhizoma powder, with no calcium oxalate needle, oil cell, brown pigment and stone cell. Microscopic identification could be made for Notoginseng Radix et Rhizoma powder and 4 common counterfeits. <b>Conclusion::Photoshop image processing software measures the diameter of the microspheres with a high speed, high accuracy, simplicity, and low requirements for measuring instruments. It provides a new way to quickly measure the diameter of microspheres. Photoshop image processing software draws a schematic diagram of the microstructure, which is convenient and faster. The original morphology of the ground-reactive microstructure provides a new method for the microstructural drawing.

ABSTRACT

OBJECTIVE@#To explore the efficacy and safety of allogeneic hematopoietic stem cell transplantation (allo-HSCT) in the treatment of relapsed or refractory peripheral T-cell lymphoma(PTCL).@*METHODS@#The clinical data of 6 patients with relapsed or refractory PTCL undergoing allo-HSCT from Sep. 2014 to Sep. 2018 in the department of hematology, aerospace center hospital were retrospectively analyzed. Complications and disease-free survival after HSCT were observed.@*RESULTS@#All the patients could well tolerate the conditioning regimen and acquired hematopoietic recon-struction. Following up till December 2018, with a median time of 11.5 months (1-51); acute GVHD developed in 2 cases and chronic GVHD developed in 5 cases, Among 6 cases one case died of viral pheumonia and the other 5 patients remained disease-free survival. The longest disease-free survival time has reached 51 months.@*CONCLUSION@#allo-HSCT is a safe and effective method for relapsed or refractory peripheral T-cell lymphoma, which can be chosen as salvage treatment method for patients with primary resistance. Optimization of the conditioning regimen may result in better efficacy of allo-HSCT.

ABSTRACT

<p><b>Objective</b>To screen out an effective substitute in the prescription of Shengjing Capsules (SJC), observe the effects of the redeveloped New SJC (NSJC) with cordyceps cephalosporium mycelia (CCM) substituted for the ingredient cordyceps sinensis in the treatment of spermatogenesis impairment (SI), and provide some experimental evidence for its application in the treatment of male infertility and sexual dysfunction.</p><p><b>METHODS</b>We equally randomized 192 male mice into 16 groups: normal saline control, SI model, high-, medium- and low-dose fermented cordycepin powder (FCP, 1.60, 0.80 and 0.40 g/kg), high-, medium- and low-dose CCM (1.60, 0.80 and 0.40 g/kg), high-, medium- and low-dose cordyceps mortierella mycelia (CMM, 1.60, 0.80 and 0.40 g/kg), high-, medium- and low-dose fermented cordyceps sinensis (FCS, 1.60, 0.80 and 0.40 g/kg), SJC (0.80 g/kg), and vitamin E (VE, 0.25 g/kg), with the SI model established in all the mice and the normal controls injected intraperitoneally with cyclophosphamide at 60 mg/kg qd for 5 consecutive days. After intragastrical medication with respective drugs, we obtained the body mass index (BMI), sexual organ coefficient, sperm count, sperm motility, and percentage of morphologically abnormal sperm (MAS) of the mice. We also randomly divided 70 male rats into 7 groups of equal number: normal control, SI model, high-, medium- and low-dose NSJC (1.12, 0.56 and 0.28 g/kg), SJC (0.56 g/kg), and VE (0.18 g/kg), the SI model constructed in the latter 6 groups of rats by gavage of adenine at 200 mg/kg qd for 5 consecutive days. After intragastrical medication with respective drugs, we examined the BMI, coefficients of sexual and renal organs, levels of reproductive hormones, testicular morphology, and fertility of the animals.</p><p><b>RESULTS</b>After medication, the mice in different groups showed different degrees of improvement in the cyclophosphamide-induced slow growth, significant increases in the testicular and epididymal coefficients, sperm count, motility and viability (P < 0.05 or P < 0.01), and a remarkable reduction in the percentage of MAS (P < 0.05 or P < 0.01). The effect was particularly significant in the CCM group and therefore CCM was chosen as the best substitute ingredient in the redeveloped NSJC. Compared with the rats in other groups, those treated with NSJC exhibited significant increases in the BMI, coefficients of sexual and renal organs and levels of serum follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone (T) and estradiol (E2) (P < 0.05 or P < 0.01), improvement of the pathologically damaged testicular morphology, elevation of the pregnancy rate and litter size, and recovery from adenine-induced SI.</p><p><b>CONCLUSIONS</b>The redeveloped New Shengjing Capsules with cordyceps cephalosporium mycelia substituted for the ingredient cordyceps sinensis can improve fertility and reverse spermatogenesis impairment in male rats. The new prescription may also be applied to the clinical treatment of male infertility and sexual dysfunction.</p>

ABSTRACT

The aim of this study was to analyze the risk factors of cytomegalovirus (CMV) infection and CMV disease after nonmyeloablative allogeneic hematopoietic stem cell transplantation(NST) and develop a rational strategy for the diagnosis, monitoring and preemptive treatment of CMV infection. The Clinical data of 80 patients undergoing NST from November 2009 to November 2012 in the hospital 307 were retrospectively analyzed. The cytomegalovirus load in peripheral blood of patients was detected by using RT-PCR. The results indicated that the incidence of CMV infection was 77.5% (62/80), and the median time for the positive CMV-DNA firstly detected by RT-PCR was day 35 (17-133) after NST. The total of 100-day cumulative incidence of CMV disease was 11.3%(9/80) after early preemptive therapy. Both univariate and multivariate analysis showed thymoglobulin (ATG) used in preconditioning regimen, other herpesvirus infection and fungal infection in medical history before NST were the risk factors of CMV infection after NST.Univariate analysis revealed that CMV viremia and ATG used in preconditioning regimen were the risk factors for CMV disease, while the same result was not found in the multivariate analysis.The incidence of CMV infection in patients with II-IV grade of aGVHD was 91.3%,while the incidence of CMV infection in patients with 0-1 grade of aGVHD was 71.9% (P = 0.06), it seems that II-IV grade of aGVHD was not the risk factor of CMV infection for NST. It is concluded that the ATG used in preconditioning regimen may increase the incidence of both CMV infection and CMV disease after NST. CMV infection easily accompanies by other herpesvirus infection and fungal infection.Therefore other herpesvirus infection and fungal infection should be attentively monitored and prevented after trans-plantation.

ABSTRACT

Objective To evaluate 18F-N- succinimidyl -4-fluorobenzoate (SFB)-Annexin B1 in detectingin vitro andin vivo apoptosis. Methods Anti-Fas antibody was used to induce apoptosis in Jurkat cells. Apoptosis in Jurkat cells was confirmed by flow cytometer (FCM). Unilateral renal ischemia/reperfusion injury was induced by transient (45 min) ligation of the renal artery in the rabbit. The rabbit was then administrated with 18F-SFB-Annexin B1 intravenously 24 h later and then imaged by PET/CT at 10,30,60,90,120 and 240 min postinjection. Apoptosis in kidney was confirmed by terminal deoxynucleotidyl transferase mediated dUTP biotin nick end labeling (TUNEL) assay and HE staining. Results The apoptosis rate induced by anti-Fas antibody was 25.98%(120 min) while that in the control group was only 1.81%. The uptake of 18F-SFB-Annexin B1in apoptosis group was greater than that in the control group. PET/CT images at 240 min showed higher uptake in the ligated kidney than the non-ligated kidney. TUNEL assay and HE staining confirmed great amount apoptotic cells in the ligated kidney. Conclusion 18 F-SFB-Annexin B1may be potentially useful in detecting apoptosis both in vitro and in vivo.

ABSTRACT

Objective To synthesize 16α-[18F]fluoro-17β-estrogen(18F-FES) with high chemical and radiochemical purity using multifunctional radiolabeling module of Explora GN and separation module of Explora LC (Explora GN/LC dual module). Methods Explora GN/LC dual module and one-pot radiochemical process were applied for fully automated synthesis and separation of 18F-FES. The radiochemical purity of 18F-FES was analyzed by HPLC. The saturation binding test of 18F-FES with ER (ER-positive MCF-7 cell line) was applied to examine the biological activity of 18F-FES. Results 18F-FES synthesis procedure took 60 min, with radiochemical yield about 45% and radiochemical purity higher than 98%. The Kd and Bmax of MCF-7 cells with 18F-FES were (2.922±0.619) nmol/L and (4.193±0.360) nmol/L respectively. Conclusions 18F-FES can be automatically synthesized by Explora GN/LC dual module with high chemical purity and radiochemical purity. It would be a potential PET tracer to be used in estrogen-related molecular imaging.

ABSTRACT

Objective To synthesize and analyze sodium 18F-fluoroacetate (FAC) and its precursor ethyl O-mesylglycolate (EOMG). Methods EOMG was synthesized using modified method. Its chemical purity was checked by HPLC and its structure was elucidated on the basis of spectral analyses. Final product of 18F-FAC was synthesized based on general nucleophilic reactions module Explora GN using EOMG as a precursor. Liquid chromatography Explora LC was applied to get rid of its chemical impurities.Then HPLC and radio-thin layer chromatography were used to assay its radiochemical purity, chemical purity and specific activity. Results EOMG was synthesized and identified. Its yield was 70% and its chemical purity was 97.0% (calculated by chromatographic peak area). The radiochemical purity of 18F-FAC was more than 98%, and its specific activity was 236. 5 MBq/μmol. Conclusion This synthetic method for 18F-FAC and its precursor can be defined as effective and highly quality-controlled.

ABSTRACT

Objective To monitor the prevalence of Keshan disease in Huangling County, Shaanxi Previnee in 2007. Methods All local residents in surveinance area of Keshan disease in Diantou and Yaoping Township of Huangling County, were surveyed and given overall examinations by electrocardiography. The patients of latent and chronic Keshan disease were examined by X-ray. The selenium contents in blood and hair were monitored by fluorospectrophotometry in surveillance and non-surveillance areas. Results No new ease of acute or sub-acute patient was found. Latent and chronic patients detected accounted for 3.92% (35/893). Only 1 ease of new latent Keshan disease was newly found. Major feature of electrocardiographic abnormality was ST-T change. By X-ray, there were 27 eases with a normal heart, 20 eases with mild enlargement, 20 eases with medium enlargement, and 16 eases with marked enlargement. Serological selenium eoncentrations were (38.67±19.58), (48.55±22.36), (67.29±17.32)μg/L, respectively in patients, internal and external eontrols(F=16.291 ,P<0.01). Selenium contents of children's hair were (0.376±0.101), (0.372±0.085), (0.436±0.085)μg/L, respectively(F= 17.032, P<0.01). Conclusions Cases of diagnosed Keshan disease were decreased compared with the previous years. There were new eases of latent Keshan disease in Huangling County, Shaanxi Province.

ABSTRACT

<p><b>OBJECTIVE</b>For anti-inflammation, relieving pain and decreasing acute toxicity, the best condition for extracting procedure of Psammosilene tunicoides was chosen. And the relationships between several solvent extracts and pharmacodynamic index were studied.</p><p><b>METHOD</b>Uniform design with multi-targets was used in the optimization process, and the evaluate index were the results of anti-inflammation, relieving pain and LD50 were employed as the evaluating indexes. At the same time, the contents of the extracts, water extract, acetic ester extract and ether extract were determined.</p><p><b>RESULT</b>The best condition is that the medicinal material is optimized with 8 times of water (pH 6-7) and 2 h of each time. The best condition is satisfactory by test verification. And the pharmacodynamic actions of anti-inflammation, relieving pain and LD, were not obviously related to the dry extracts, water extract, acetic ester extract and ether extract.</p><p><b>CONCLUSION</b>Uniform design method with pharmacodynamic index has the merit of being relaated to the function and symptom directly. The extracted twice technology condition optimized is stable, feasible and suitable for industry production. And it is not suitable to take the solvent extracts as quality control index related to the function and symptom.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the transdermal osmosis process of Aconitum brachypodum's liniment, gel and patcher to provide basis for selecting dosage form and controlling the quality.</p><p><b>METHOD</b>Taking the cumulate rate of transdermal as index, a imitated Fick's diffusion device was used for the investigating the transdermal osmosis course of the three preparations. The best transdermal mathematics models are obtained and the relations between the transdermal course and the release course are analysed.</p><p><b>RESULT</b>The three preparations have different characteristics of transdermal osmosis course. The liniment meets dynamics 0 order process, the gel and the patcher meet dynamic 0 order process of non-corroded drug system. And the relation is good cubic equation between their transdermal course and release course.</p><p><b>CONCLUSION</b>The transdermal osmosis experiment in vitro for three preparations can provide basis for selecting dosage form and the quality control in future studies.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate a possibility of repairing damaged brain by intracerebroventricular transplantation of neural stem cells (NSCs) in the adult mice subjected to glutamate-induced excitotoxic injury.</p><p><b>METHODS</b>Mouse NSCs were isolated from the brains of embryos at 15-day postcoitum (dpc). The expression of nestin, a special antigen for NSC, was detected by immunocytochemistry. Immunofluorescence staining was carried out to observe the survival and location of transplanted NSCs. The animals in the MSG + NSCs group received intracerebroventricular transplantation of NSCs (approximately 1.0 x 10(5) cells) separately on day 1 and day 10 after 10-d MSG exposure (4.0 g/kg per day). The mice in control and MSG groups received intracerebroventricular injection of Dulbecco's minimum essential medium (DMEM) instead of NSCs. On day 11 after the last NSC transplantation, the test of Y-maze discrimination learning was performed, and then the histopathology of the animal brains was studied to analyze the MSG-induced functional and morphological changes of brain and the effects of intracerebroventricular transplantation of NSCs on the brain repair.</p><p><b>RESULTS</b>The isolated cells were Nestin-positive. The grafted NSCs in the host brain were region-specifically survived at 10-d post-transplantation. Intracerebroventricular transplantation of NSCs obviously facilitated the brain recovery from glutamate-induced behavioral disturbances and histopathological impairs in adult mice.</p><p><b>CONCLUSION</b>Intracerebroventricular transplantation of NSCs may be feasible in repairing diseased or damaged brain tissue.</p>

Objective To analyse the femoral head coverage rate of CT in evaluation of Salter osteotomy for developmental dislocation of hip(DDH) in children. Methods Thirty-eight patients with unilateral DDH for Salter osteotomy were enrolled,and X ray photography of pelvis and spiral CT scanning of femoral articulation were performed one week before and 6 months after Salter osteotomy.The femoral head coverage rate of X ray and that of CT were calculated and compared.Another 38 children with normal femoral articulation were served as controls.
